LB7 tuning questions

I'm wanting to do a little tuning on my 01 LB7. Can I read it in vehicle with my EFI or do I need to do it with a bench flasher? Or should I just find a later ECU and flash it with a 6006 OS?

You can read it out with the ecm in the truck still.
