no, but they are "supposed" to go back in in the same orientation as they were found. I haven't seen it affect anything yet, but that's not to say that it can't.

Correct.
The company that makes the gears tests them for noise after every run to see which orientation is the quietest and that's how the gears are installed at the factory. The marks might be up or down or one up and another down.
I set all of mine with the marks up just because I have been doing it for over 1600 Allisons and no other reason. It doesn't matter.
